This genomic element is located in the intergenic region between the FAM174B (family with sequence similarity 174 member B) and LINC01578 (long intergenic non-protein coding RNA 1578) genes. It was defined as a high expression actively transcribed enhancer based on the presence of balanced bidirectional capped transcripts by cap analysis of gene expression (CAGE). It was validated as a functional enhancer by reporter assays in both HeLa and HepG2 cells. A sub-region was also validated as a functional enhancer by high-throughput massively parallel reporter assays (MPRAs) in HepG2 cells. This sub-region contains a conserved motif for the ZFP161 transcription factor, and mutation of the motif results in reduced enhancer activity. Another sub-region was further validated as a functional enhancer by the Sharpr-MPRA technique (Systematic high-resolution activation and repression profiling with reporter tiling using MPRAs) in HepG2 cells (group: HepG2 Activating DNase unmatched - State 1:Tss, active promoter, TSS/CpG island region).
