This gene encodes a member of the DnaJ or Hsp40 (heat shock protein 40 kD) family of proteins. DNAJ family members are characterized by a highly conserved amino acid stretch called the 'J-domain' and function as one of the two major classes of molecular chaperones involved in a wide range of cellular events, such as protein folding and oligomeric protein complex assembly. The encoded protein is a molecular chaperone that stimulates the ATPase activity of Hsp70 heat-shock proteins in order to promote protein folding and prevent misfolded protein aggregation. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Sep 2015]
GeneCards Summary for DNAJB1 Gene
DNAJB1 (DnaJ Heat Shock Protein Family (Hsp40) Member B1) is a Protein Coding gene. Diseases associated with DNAJB1 include Fibrolamellar Carcinoma and Mixed Fibrolamellar Hepatocellular Carcinoma. Among its related pathways are Signaling by GPCR and Cellular response to heat stress. Gene Ontology (GO) annotations related to this gene include unfolded protein binding and ATPase binding. An important paralog of this gene is DNAJB4.
UniProtKB/Swiss-Prot Summary for DNAJB1 Gene
Interacts with HSP70 and can stimulate its ATPase activity. Stimulates the association between HSC70 and HIP. Negatively regulates heat shock-induced HSF1 transcriptional activity during the attenuation and recovery phase period of the heat shock response (PubMed:9499401). Stimulates ATP hydrolysis and the folding of unfolded proteins mediated by HSPA1A/B (in vitro) (PubMed:24318877).
