Summaries for CD44 Gene

Entrez Gene Summary for CD44 Gene

  • The protein encoded by this gene is a cell-surface glycoprotein involved in cell-cell interactions, cell adhesion and migration. It is a receptor for hyaluronic acid (HA) and can also interact with other ligands, such as osteopontin, collagens, and matrix metalloproteinases (MMPs). This protein participates in a wide variety of cellular functions including lymphocyte activation, recirculation and homing, hematopoiesis, and tumor metastasis. Transcripts for this gene undergo complex alternative splicing that results in many functionally distinct isoforms, however, the full length nature of some of these variants has not been determined. Alternative splicing is the basis for the structural and functional diversity of this protein, and may be related to tumor metastasis. [provided by RefSeq, Jul 2008]

GeneCards Summary for CD44 Gene

CD44 (CD44 Molecule (Indian Blood Group)) is a Protein Coding gene. Diseases associated with CD44 include Superficial Keratitis and Lichen Sclerosus. Among its related pathways are Cell adhesion_ECM remodeling and G-protein signaling G-Protein alpha-i signaling cascades. Gene Ontology (GO) annotations related to this gene include transmembrane signaling receptor activity and cytokine receptor activity. An important paralog of this gene is LYVE1.

UniProtKB/Swiss-Prot for CD44 Gene

  • Receptor for hyaluronic acid (HA). Mediates cell-cell and cell-matrix interactions through its affinity for HA, and possibly also through its affinity for other ligands such as osteopontin, collagens, and matrix metalloproteinases (MMPs). Adhesion with HA plays an important role in cell migration, tumor growth and progression. In cancer cells, may play an important role in invadopodia formation. Also involved in lymphocyte activation, recirculation and homing, and in hematopoiesis. Altered expression or dysfunction causes numerous pathogenic phenotypes. Great protein heterogeneity due to numerous alternative splicing and post-translational modification events. Receptor for LGALS9; the interaction enhances binding of SMAD3 to the FOXP3 promoter, leading to up-regulation of FOXP3 expression and increased induced regulatory T (iTreg) cell stability and suppressive function (By similarity).

Post-translational modifications for CD44 Gene

  • Proteolytically cleaved in the extracellular matrix by specific proteinases (possibly MMPs) in several cell lines and tumors.
  • N- and O-glycosylated. O-glycosylation contains more-or-less-sulfated chondroitin sulfate glycans, whose number may affect the accessibility of specific proteinases to their cleavage site(s). It is uncertain if O-glycosylation occurs on Thr-637 or Thr-638.
  • Phosphorylated; activation of PKC results in the dephosphorylation of Ser-706 (constitutive phosphorylation site), and the phosphorylation of Ser-672.
  • Glycosylation at Thr638, Thr637, posLast=636636, posLast=599599, posLast=578578, posLast=572572, Ser570, Thr567, Ser562, Thr561, posLast=560560, Asn548, posLast=541541, posLast=514514, Thr484, Thr480, Ser458, Ser434, posLast=433433, posLast=430430, Thr426, posLast=424424, Thr423, posLast=422422, posLast=350350, Ser330, Thr325, posLast=318318, posLast=200200, posLast=174174, posLast=120120, Asn110, Asn100, Asn57, and Asn25
