The encoded protein is a component of a largemultimeric complex, termed the retromer complex, involved in retrograde transport of proteins from endosomes to thetrans-Golgi network. The close structural similarity between the yeast and human proteins that make up this complexsuggests a similarity in function. Expression studies in yeast and mammalian cells indicate that this proteininteracts directly with VPS35, which serves as the core of the retromer complex. Defects in VPS35 are the cause of Parkinson disease type 17 (PARK17) [MIM:614203]. PARK17 is an autosomaldominant, adult-onset form of Parkinson disease. Parkinson disease is a complex neurodegenerative disordercharacterized by bradykinesia, resting tremor, muscular rigidity and postural instability, as well as by a clinicallysignificant response to treatment with levodopa.
