About This Section Entrez Gene summary for MMP9 : Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The enzyme encoded by this gene degrades type IV and V collagens. Studies in rhesus monkeys suggest that the enzyme is involved in IL-8-induced mobilization of hematopoietic progenitor cells from bone marrow, and murine studies suggest a role in tumor-associated tissue remodeling. (provided by RefSeq, Jul 2008) UniProtKB/Swiss-Prot: MMP9_HUMAN, P14780 Function : May play an essential role in local proteolysis of the extracellular matrix and in leukocyte migration. Couldplay a role in bone osteoclastic resorption. Cleaves KiSS1 at a Gly- -Leu bond. Cleaves type IV and type V collagen into large C-terminal three quarter fragments and shorter N-terminal one quarter fragments. Degrades fibronectin but not laminin or Pz-peptide summary
for MMP9 : Matrix metalloproteases (matrix metalloproteinase, MMPs), also called matrixins, are zinc-dependentendopeptidases that are the major proteases involved in ECM degradation. MMPs are capable of degrading awide range of extracellular molecules and a number of bioactive molecules. 24 matrixin genes have beenidentified in humans, which can be organized into six groups based on domain organization and substratepreference: Collagenases (MMP-1, -8 and -13), Gelatinases (MMP-2 and MMP-9), Stromelysins (MMP-3, -10 and-11), Matrilysin (MMP-7 and MMP-26), Membrane-type (MT)-MMPs (MMP-14, -15, -16, -17, -24 and -25) and others(MMP-12, -19, -20, -21, -23, -27 and -28). MMP activity is regulated by two major endogenous inhibitors:alpha2-macroglobulin and tissue inhibitors of metalloproteases (TIMPs). MMPs play a central role in cellproliferation, migration, differentiation, angiogenesis, apoptosis and host defences. Dysregulatoin of MMPshas been implicated in many diseases including arthritis, chronic ulcers, encephalomyelitis and cancer.Tumour metastasis is a multistep process involving the dessemination of tumor cells from the primary tumorto secondarys at a distant organ or tissue. One of the first steps in metastasis is the degradation of thebasement membrane, a process in which MMPs have been implicated. MMPs are secreted by tumor cells themselvesor by surrounding stromal cells stimulated by the nearby tumor. Numerous studies have linked altered MMPexpression in different human cancers with poor disease prognosis. MMP-1, -2, -3, -7, -9, -13 and -14 allhave elevated expression in primary tumors and/or metastases. Synthetic or natural inhibitors of MMPs resultin inhibition of metastasis, while up-regulation of MMPs led to enhanced cancer cell invasion. Gene Wiki entry for MMP9

About This Section
MMP9 for disorders About GeneDecksing OMIM gene information: 120361 OMIM disorders : 613073 UniProtKB/Swiss-Prot: MMP9_HUMAN, P14780
Defects in MMP9 may be a cause of susceptibility to intervertebral disc disease (IDD) [MIM:603932]; also known as lumbar disk herniation (LDH). IDD is one of the most common musculo-skeletal disorders and the predominant cause of low-back pain and unilateral leg pain Defects in MMP9 are the cause of metaphyseal anadysplasia type 2 (MANDP2) [MIM:613073]. Metaphyseal anadysplasia consists of an abnormal bone development characterized by severe skeletal changes that, in contrast with the progressive course of most other skeletal dysplasias, resolve spontaneously with age. Clinical characteristics are evident from the first months of life and include slight shortness of stature and a mild varus deformity of the legs. Patients attain a normal stature in adolescence and show improvement or complete resolution of varus deformity of the legs and rhizomelic micromelia 20/417 diseases for MMP9 (see all 417 ): About MalaCards angle-closure glaucoma metaphyseal anadysplasia arthritis primary angle-closure glaucoma metaphyseal anadysplasia 2 spinal cord injury internal hemorrhoid relapsing-remitting multiple sclerosis hyper-ige syndrome pigmented villonodular synovitis toxic epidermal necrolysis encephalomyelitis sorsby fundus dystrophy lentigo maligna melanoma open-angle glaucoma otitis media hepatocellular carcinoma (fibrolamellar variant) stevens-johnson syndrome villonodular synovitis middle ear cholesteatoma 20/27 diseases from the University of Copenhagen DISEASES database for MMP9 (see all 27 ):Carcinoma Atherosclerosis Heart disease Cerebrovascular accident Abdominal aortic aneurysm Breast cancer Arthritis Lung cancer Brain edema Chronic obstructive pulmonary disease Malignant glioma Periodontitis Colorectal cancer Asthma Prostate cancer Hepatocellular carcinoma Multiple sclerosis Hypertension Ovarian cancer Pancreatic cancer 10/96 Novoseek disease relationships for MMP9 gene (see all 96 ) About this table
