This gene encodes the D2 subtype of the dopamine receptor. This G-protein coupled receptor inhibits adenylyl cyclase activity. A missense mutation in this gene causes myoclonus dystonia; other mutations have been associated with schizophrenia. Alternative splicing of this gene results in two transcript variants encoding different isoforms. D2 receptors are members of the dopamine receptor G-protein-coupled receptor family that also includes D1, D3, D4 and D5. They are located primarily in the caudate putamen, nucleus accumbens and olfactory tubercle where they are involved in the modulation of locomotion, reward, reinforcement and memory and learning. The human D2 receptor gene has been localized to chromosome 11 (11q22-23).
