DNAJ proteins may have up to 3 distinct domains: a conserved 70-amino acid Jdomain, usually at the N terminus; a glycine/phenylalanine (G/F)-rich region; and a C-terminal cysteine-rich region(Ohtsuka and Hata, 2000 (PubMed 11147971)).(supplied by OMIM, Mar 2008) UniProtKB/Swiss-Prot: DJB11_HUMAN, Q9UBS4Function: Serves as a co-chaperone for HSPA5. Binds todenatured substrates in an ATP-independent manner. Interacts via the J domain with HSPA5 in an ATP-dependent manner
Subcellular location: Endoplasmic reticulum lumen. Note=Associated with the ER membrane in a C-terminallyepitope-tagged construct
Caution: PubMed:11584023 reported a cytosolic, as well as nuclear subcellular location. This result was obtained usingan N-terminally GFP-tagged construct which most probably affected signal peptide-driven targeting to the ER. As aconsequence, the in vivo revelance of the observed interaction with APOBEC1, a nuclear protein, is dubious. This holdstrue for the interaction with PWP1
